No water in the toilet

Hi, my toilet want fill up with water, from reading various posts, i think the problem is the inlet value probably needs replaced from this thread (http://www.diynot.com/diy/threads/toilet-slow-to-fill-up-cistern-after-flushing.202360/page-2). I have got the part but i cant work out how to get the bolt off to replace it.....i have tried turning left/right, tried using those grip things from the kitchen. Do i need a tool to open it and which direction should i be doing it in?

One last question, for turning the water off on the toilet, i have found a screw thing on the pipe, i think i turn it clockwise to turn off, is that right?

I wouldn't mess around with that. Go and get a fluid master pro fill valve. Comes with a nice brass thread on the bottom. Remove that thing and replace. Use a sponge to get rid of the water in the bottom of the cistern prior to replacing. Check the water is off before you start.
 
The screw you talk about should be horizontal or across the pipe to turn off. In line means it's on.
